ADAPTOL General Description: International Nonproprietary Name: 2,4,6,8-tetramethyl-2,4,6,8-tetraazabicyclo(3.3.0)octanedione-3,7; Main physicochemical properties: White or almost white, round, flat, uncoated tablets with a bevel and score; Composition: 1 uncoated tablet contains: Mebicar – 300 mg or 500 mg. Excipients: Methylcellulose, Calcium stearate. Dosage form: Uncoated tablets, 300 mg or 500 mg. Pharmacological group: Anxiolytic agent (tranquilizer). ATC code: N05B. See blog: Adaptol — a preparation for relieving neuroses and neurosis-like conditions. Pharmacological properties: Adaptol-Aversi is an anxiolytic agent that acts on the activity of structures within the limbic-reticular complex, specifically on the emotional zones of the hypothalamus. It balances neuromediator systems (gamma-aminobutyric acid, cholinergic, serotonergic, and adrenergic), promotes the integration of central nervous system functions, including mental functions, and normalizes emotional state. The preparation relieves or weakens restlessness, anxiety, fear, internal emotional tension, and irritability. The sedative effect of Adaptol-Aversi does not cause muscle relaxation or impaired motor coordination. The preparation can be used during the workday ("daytime tranquilizer") as it does not cause a decrease in mental or physical activity. The preparation does not have a hypnotic effect, but it enhances the action of hypnotics and regulates sleep in case of sleep disturbances. Euphoria or elevated mood is not observed during the use of the preparation. Adaptol-Aversi relieves or alleviates nicotine withdrawal symptoms. In addition to its sedative effect, the preparation exhibits nootropic action. Clinical studies have shown that the active ingredient of the preparation increases the speed and logic of thinking, promotes concentration, and increases mental work capacity. At the same time, it does not stimulate the symptomatology of productive psychopathological disorders (delusions, pathological emotional activity). Adaptol-Aversi has antioxidant properties, therefore, during oxidative stress of various origins, it acts as a membrane-stabilizing, adaptogenic, and cerebroprotective agent. Pharmacokinetics: Adaptol-Aversi is well absorbed from the gastrointestinal tract. Bioavailability is 70-80%. 40% of the administered dose binds to erythrocytes. The remaining portion is in the plasma in free form, which allows the active substance of the preparation to spread unimpeded throughout the body and freely cross cell membranes. Maximum concentration (Cmax) in blood plasma is reached after 30 minutes and its high level is maintained for 3-4 hours, after which it gradually decreases. 55-70% of the administered dose is excreted with urine, the rest – unchanged with feces within 24 hours. The active substance of the preparation is not metabolized or accumulated in the body. Indications: • Neuroses and neurosis-like conditions accompanied by irritability, emotional instability, anxiety, and fear; • Improvement of tolerance to neuroleptics and tranquilizers by alleviating their somatic-vegetative and neurological side effects; • Cardialgia of various origins (not associated with ischemic heart disease); • Nicotine dependence (as part of complex treatment). Dosage and administration: Adaptol-Aversi is taken orally at 300-500 mg 2-3 times a day. The maximum single dose is 3 g, the maximum daily dose is 10 g. The duration of the treatment course ranges from several days to 2-3 months. For the treatment of nicotine dependence, it is prescribed at 600-900 mg 3 times a day for 5-6 weeks. Tolerance and withdrawal syndrome do not develop with Adaptol-Aversi. The intake of the preparation is not dependent on food intake. If a scheduled dose is missed, treatment should be continued with the previously prescribed dose, without taking an additional dose. Medical consultation is required if necessary. Side effects: Adaptol-Aversi is characterized by good tolerability. When taking high doses of the preparation, allergic reactions (e.g., skin itching) or dyspeptic events may occur. In this case, it is necessary to discontinue the preparation. Short-term decrease in blood pressure and hypothermia may occur, which normalize on their own, as well as weakness and dizziness. Contraindications: Hypersensitivity to Mebicar or other components of the preparation. Interactions with other drugs: Adaptol-Aversi can be used with other tranquilizers (benzodiazepine group), antidepressants, psychostimulants, hypnotics, and neuroleptics. Overdose: Adaptol-Aversi is a low-toxicity preparation. In case of overdose, detoxification therapy is carried out. Pregnancy and lactation: Due to the lack of adequate clinical studies, the use of Adaptol-Aversi during pregnancy and breastfeeding is not recommended. Special precautions: No special indications. Effect on ability to drive and operate machinery: No effect on the ability to drive vehicles and operate other machinery. Packaging: 10 tablets in a contour cell package, 2 contour packages in a cardboard box. Storage conditions: Store at a temperature of 15-25ºC, in a dry place, protected from children. Shelf life: 4 years. The preparation is not allowed to be used after the expiry date indicated on the packaging. Pharmacy dispensing condition: Prescription only. Manufacturer: LLC "Aversi-Rational" (Georgia).
